Thermo Scientific™ Easy-Titer Antibody Assay Kits enable accurate determination of antibody concentration in serum and culture supernatant, and include kits specific for mouse IgG, rabbit IgG, human IgG (H+L), human IgG (Fc), or human IgM.
nThese kits include antibody-sensitized microspheres to measure the specific concentration of mouse, rabbit and human antibodies by an easy and rapid micro-agglutination technique using standard microplates and UV-Vis plate reader (spectrophotometer). Each kit is specific for a particular species and class of immunoglobulin and, unlike total protein assays, can specifically measure the concentration of target antibody in samples (e.g., serum, plasma, culture supernatant) that contain other proteins. The kits are extremely sensitive, requiring very small sample volumes. Antibody concentration is determined from the assay response (absorbance) by comparison to a standard curve prepared using dilutions of a known antibody sample (sold separately).

Perform a variety of antibody affinity purification methods with Thermo Scientific™ Pierce Protein A Agarose, a standard-capacity Protein A beaded agarose resin.nn nnPierce Protein A Agarose consists of purified native Protein A that has been covalently immobilized onto high-quality crosslinked 6% beaded agarose. The agarose beads have physical and chemical properties that enable them to be used in a variety of batch- or column-type affinity purification procedure. Several package formats are offered: 5mL and 25mL bottles of resin slurry, 1mL pre-packed columns and a complete IgG purification kit.nnProtein A is a bacteria-derived protein that binds with high affinity and specificity to the Fc portion of antibodies, especially those of the IgG class. Compared to its alternative (Protein G), Protein A provides the highest binding capacity for subclasses of IgG from rabbits, pigs, dogs and cats. Protein A also has higher binding capacity for human IgG, except for IgG3, which it binds weakly. Protein A also binds weakly to mouse IgG1 and is not recommended for most applications with that antibody isotype.

Thermo Scientific™ 1-Step™ Ultra TMB-ELISA Substrate Solution SKU: 34028
Thermo Scientific™ Pierce TMB ELISA Substrates detect horseradish peroxidase (HRP) activity, yielding a blue color (Amax = 370nm and 652nm) that changes to yellow (Amax = 450nm) upon addition of a sulfuric or phosphoric acid stop solution.
nTMB (3,3′,5,5′ tetramethylbenzidine) is a chromagen that yields a blue color when oxidized with hydrogen peroxide (catalyzed by HRP) with major absorbances at 370 and 652nm. The color then changes to yellow (measured at 450nm) upon addition of sulfuric or phosphoric acid to stop the reaction. TMB is very sensitive and more quickly oxidized than other HRP substrates, resulting in faster color development. Unlike other commercially available substrates, these products contain no DMF or DMSO. There are three formulations that differ primarily in their sensitivities. Slow TMB is intermediate in sensitivity, ideal for kinetic readings. The sensitivity of the Turbo TMB compares to that of OPD used at approximately 1mg/mL. Ultra TMB-ELISA produces the highest signal-to-noise ratio and sensitivity in the picogram range. Highlights:

This reducing agent-compatible (RAC) version of the Thermo Scientific™ Pierce BCA Protein Assay provides accurate determination of protein concentration in samples that contain dithiothreitol (DTT), 2-mercaptoethanol (BME), TCEP and other disulfide reducing agents.
The BCA Protein Assay Kit – Reducing Agent Compatible (BCA-RAC) provides all of the advantages of the original BCA Assay, plus compatibility with disulfide reducing agents at concentrations routinely used in protein sample buffers. This special adaptation of the popular Pierce BCA Protein Assay method enables accurate protein concentration measurement for samples containing DTT, 2-ME or TCEP. Available in tube and microplate formats, these reducing agent compatible (RAC) BCA Kits extend the already broad reagent compatibility of the BCA Protein Assay protocol to include nearly all types of components commonly present in protein research samples.

Thermo Scientific™ ActivX™ Desthiobiotin-GTP Probe X
ActivX™ Desthiobiotin-GTP Probes contain a modified biotin attached to the nucleotide by a labile acyl-phosphate bond. ActivX active-site enrichment probes are advantageous for determining active enzyme levels through the use of analog-tagged probes to allow profiling of both inactive and active enzymes. The ActivX probes selectively enrich only those enzymes that are functionally active and biologically relevant at the time of labeling. With selective enrichment, one can identify and profile target enzyme classes across samples or can assess the specificity and affinity of enzyme inhibitors.
After removal of GTP or GDP nucleotides from enzymes, the desthiobiotin-GTP probe can be used to covalently modify conserved lysine residues in the GTPase nucleotide-binding site. Desthiobiotin-GTP can selectively enrich, identify and profile target enzyme classes in samples. Pre-incubation of samples with small-molecule inhibitors that compete with active-site probes can be used to determine inhibitor binding affinity and target specificity. The ActivX Desthiobiotin-GTP Probe covalently labels the active site of GTPases and GTPase subunits of G-protein coupled receptors.
Applications of ActivX Desthiobiotin-GTP Probe:
• Broad enrichment of GTP-binding proteins from tissues, cells and sub-cellular proteomes
• Enrichment of enzymes based on function
• Profiling of dozens to hundreds of inhibitor targetsAssessment of active-site labeling with desthiobioin probes can be accomplished by either Western blot or mass spectrometry (MS). For the Western blot workflow, desthiobiotin-labeled proteins are enriched for SDS-PAGE analysis and subsequent detection with specific antibodies. For the MS workflow, desthiobiotin-labeled proteins are reduced, alkylated and enzymatically digested to peptides. Only the desthiobiotin-labeled, active-site peptides are enriched for analysis by LC-MS/MS. Both workflows can be used for determining inhibitor target binding, but only the MS workflow can identify global inhibitor targets and off-targets.
