Thermo Scientific™ Easy-Titer Antibody Assay Kits enable accurate determination of antibody concentration in serum and culture supernatant, and include kits specific for mouse IgG, rabbit IgG, human IgG (H+L), human IgG (Fc), or human IgM.
nThese kits include antibody-sensitized microspheres to measure the specific concentration of mouse, rabbit and human antibodies by an easy and rapid micro-agglutination technique using standard microplates and UV-Vis plate reader (spectrophotometer). Each kit is specific for a particular species and class of immunoglobulin and, unlike total protein assays, can specifically measure the concentration of target antibody in samples (e.g., serum, plasma, culture supernatant) that contain other proteins. The kits are extremely sensitive, requiring very small sample volumes. Thermo Scientific™ Pierce TMB ELISA Substrates detect horseradish peroxidase (HRP) activity, yielding a blue color (Amax = 370nm and 652nm) that changes to yellow (Amax = 450nm) upon addition of a sulfuric or phosphoric acid stop solution.
nTMB (3,3′,5,5′ tetramethylbenzidine) is a chromagen that yields a blue color when oxidized with hydrogen peroxide (catalyzed by HRP) with major absorbances at 370 and 652nm. The color then changes to yellow (measured at 450nm) upon addition of sulfuric or phosphoric acid to stop the reaction. TMB is very sensitive and more quickly oxidized than other HRP substrates, resulting in faster color development. Unlike other commercially available substrates, these products contain no DMF or DMSO. There are three formulations that differ primarily in their sensitivities. Slow TMB is intermediate in sensitivity, ideal for kinetic readings. The sensitivity of the Turbo TMB compares to that of OPD used at approximately 1mg/mL. This reducing agent-compatible (RAC) version of the Thermo Scientific™ Pierce BCA Protein Assay provides accurate determination of protein concentration in samples that contain dithiothreitol (DTT), 2-mercaptoethanol (BME), TCEP and other disulfide reducing agents.

Thermo Scientific™ ActivX™ Desthiobiotin-GTP Probe X
ActivX™ Desthiobiotin-GTP Probes contain a modified biotin attached to the nucleotide by a labile acyl-phosphate bond. ActivX active-site enrichment probes are advantageous for determining active enzyme levels through the use of analog-tagged probes to allow profiling of both inactive and active enzymes. The ActivX probes selectively enrich only those enzymes that are functionally active and biologically relevant at the time of labeling. With selective enrichment, one can identify and profile target enzyme classes across samples or can assess the specificity and affinity of enzyme inhibitors.
After removal of GTP or GDP nucleotides from enzymes, the desthiobiotin-GTP probe can be used to covalently modify conserved lysine residues in the GTPase nucleotide-binding site. Desthiobiotin-GTP can selectively enrich, identify and profile target enzyme classes in samples. Pre-incubation of samples with small-molecule inhibitors that compete with active-site probes can be used to determine inhibitor binding affinity and target specificity. The ActivX Desthiobiotin-GTP Probe covalently labels the active site of GTPases and GTPase subunits of G-protein coupled receptors.

Easy-Titer Antibody Assays are faster and easier than ELISA:
1. Prepare standards (5 to 500ng/mL) by diluting purified antibody in Kit Dilution Buffer
2. Prepare samples by diluting at least 20-fold in Dilution Buffer to within assay range (8 to 500ng/mL).
3. Vortex vial of microsphere beads to create homogeneous suspension.
4. Pipette 20μL of bead suspension and 20μL of each sample and standard into 96-well microplate wells.
5. Incubate microplate for 5 minutes with vigorous mixing.
6. Add 100μL of Kit Blocking Reagent.
7. Incubate microplate for 5 minutes with vigorous mixing.
8. Measure absorbance on standard plate reader (340nm or 405nm).
9. Plot standard curve and interpolate samples to determine concentration.

3. Vortex vial of microsphere beads to create homogeneous suspension.
4. Pipette 20μL of bead suspension and 20μL of each sample and standard into 96-well microplate wells.
5. Incubate microplate for 5 minutes with vigorous mixing.
6. Add 100μL of Kit Blocking Reagent.
7. Incubate microplate for 5 minutes with vigorous mixing.

Thermo Scientific™ Pierce Glycoprotein Carbohydrate Estimation Kit enables the amount of protein glycosylation to be measured as the percent of total purified protein mass.
The Glycoprotein Carbohydrate Estimation Kit includes six purified glycoprotein standards and the required assay reagents to effectively estimate the amount of oxidizable glycosylation (% carbohydrate by weight) of purified protein samples. Sugar groups in the glycoprotein sample are first oxidized with sodium meta-periodate to produce detectable aldehyde groups.
