DYKDDDDK Epitope Tag Synthetic Peptide, PEP-087, from Invitrogen.
nDYKDDDDK Tag (FLAG-tag, FLAG octapeptide) is a polypeptide protein tag that can be added to a protein using recombinant DNA technology. The FLAG tag structure has been optimized for compatibility with the proteins it is attached to, in that it is more hydrophilic than other common epitope tags and therefore less likely to denature or inactivate proteins to which it is appended. A FLAG-tag can be used in many different assays that require recognition by an antibody. If there is no antibody against the studied protein, adding a FLAG-tag to this protein allows one to follow the protein with an antibody against the FLAG sequence. Examples are cellular localization studies by immunofluorescence or detection by SDS PAGE protein electrophoresis. Tagging with xxxDDDDK may be done at the N-terminus, N-terminus preceded by a methionine residue, C-terminus, and in internal positions of the target protein. The small size of the epitope tag and its high hydrophilicity tend to decrease the possibility of interference with protein expression, proteolytic maturation, antigenicity and function. The enterokinase cleavage site allows it to be completely removed from the purified fusion proteins. similar epitope as the FLAG tag from SigmaThis peptide may be used for neutralization and control experiments with the polyclonal antibody that reacts with this product (Product &Num; PA1-984B) and fusion proteins containing the epitope tag DYKDDDDK. Using a solution of peptide of equal volume and concentration to the corresponding antibody will yield a large molar excess of peptide (?70-fold) for competitive inhibition of antibody-protein binding reactions.Reconstitute with 0.1 mL of distilled water.

Essential 8™ Medium X
Essential 8™ Medium is a xeno-free and feeder-free medium specially formulated for the growth and expansion of human pluripotent stem cells (PSCs). Originally developed by Guokai Chen et al. in the laboratory of James Thomson (published as ‘E8’) and validated by Cellular Dynamics International, Essential 8™ Medium has been extensively tested and proved to maintain pluripotency in multiple iPSC lines.
